  • 機能

    Receptor for glucocorticoids (GC). Has a dual mode of action: as a transcription factor that binds to glucocorticoid response elements (GRE) and as a modulator of other transcription factors. Affects inflammatory responses, cellular proliferation and differentiation in target tissues. Could act as a coactivator for STAT5-dependent transcription upon growth hormone (GH) stimulation and could reveal an essential role of hepatic GR in the control of body growth. Involved in chromatin remodeling. Plays a significant role in transactivation. Involved in nuclear translocation.

    Widely expressed. In the heart, detected in left and right atria, left and right ventricles, aorta, apex, intraventricular septum, and atrioventricular node as well as whole adult and fetal heart.

    Defects in NR3C1 are a cause of glucocorticoid resistance (GCRES) [MIM:138040]; also known as cortisol resistance. It is a hypertensive, hyperandrogenic disorder characterized by increased serum cortisol concentrations. Inheritance is autosomal dominant.

    Sumoylated; this reduces transcription transactivation.
    Ubiquitinated; restricts glucocorticoid-mediated transcriptional signaling.

Samples were blocked with 10% goat serum in 0.1M PBS. Samples were incubated with the primary antibody for 48 hours at 4°C. A biotin-conjugated goat anti-rabbit was used as the secondary antibody.

    AbID: 3671, Anti-Glucocorticoid Receptor antibody Rating: Inconclusive Image: Anti-Glucocorticoid Receptor antibody for Western blot (Human) Sample: Species: Human Type: Cell lysate - whole cell Loading amount: 50 µg Specification: HELA Application: Application: Gel Running Conditions: Reduced Denaturing Blocking step: Milk as blocking agent for 1 hour at 23°C Blocking Concentration: 3% Other detail: Dilution: 1/100 Incubation time: 16 hours at 4°C Diluent: TBST Secondary Antibody: Name: Non-Abcam Antibody was used: GOAT ANTI-RABBIT Conjugation: HRP Dilution: 1/2000 Detection: Detection method: ECL Exposure: 1 minute and 0 seconds Bands: Specific: 86? kDa Non-specific: 64 kDa Additional Data: Additional Notes: A 1:1000 dilution of primary antibody gave a strong 64KD band, which the antibody also pulled during IP. Since the reported 100KD band was not detected, the membranes were incubated with a 1:100 dilution of primary to reveal a 86 KD band (which may be the receptor), along with several other nonspecific bands. It may be an issue with just this aliquot of antibody, as even ICC with this antibody resulted in nonspecific staining.

    Answer

    Thank you for submitting you abreview for ab3671. I'm sorry that it has not performed as expected with the Western blotting or the ICC. Having reviewed the protocol there may be a few things I can suggest in order to improve the results. As you have been seeing a band in the expected region at a dilution of 1/100 I would continue to try with this dilution and try to reduce the amount of background observed. This may be improved by trying BSA (3% in TBS-T) as the blocking agent, incubating for 1 hour at room temperature as you have been doing. I would also suggest diluting your antibody in 1% BSA TBS-T and try incubating at room temperature for 1 hour as well as 4°C overnight as you have been doing. If the antibody continues to produce poor results please do not hesitate to let me know. If the antibody was purchased within the last 6 months and has been used as recommended on the datasheet you would be eligible for a replacement antibody (this one or an alternative from our catalogue) or a refund.
