    Immunofluorescent staining obtained with ab16053 Rabbit polyclonal to GAP43 in rat dorsal root ganglion, 2 weeks after a spinal nerve injury. The staining is localised in the cytoplasm of some lesioned neurons (arrow in B) and in many axons (arrow heads in B). Image B (X40 objective) is a higher magnification of A (X20). The staining disappeared completely when the antibody was pre-incubated with the immunising (blocking) peptide (ab16379).

    Protocol details: Rats were intracardially perfused with 4% paraformaldehyde, DRG tissue was post-fixed overnight in the same fixative, cryoprotected in 20% sucrose and frozen in OCT. IHC free floating was performed on the fixed, frozen and cut (20µm sections) rat tissue. Primary antibody ab16053 was incubated overnight at toom temperature at 1/300. Secondary antibody Alexa fluor 488 was incubated at 1/1000 for 2 hours at room temperature.

Question

We are planing to use anti-GAP43 to label regenerating axons of the sciatic nerves in mice (immunofluorescence). Ab7462 is known to work well, but it is either discontinuous or  change to another Catalog number (?). We sincerely hope you can find a replacement for us.

We would recommend using ab16053 in place of the discontinued item. Ab16053 has been tested in and is guaranteed in IF on mouse samples. This product has also been featured in 5 publications which we are aware of and has excellent Abreviews. 3) Description of the problem

I perform the immunofluorescent stain on my cultured hippocampal neuron with this antibody, no matter fixed with 4%PFA or pure methanol, I can not get the specific stain as the product sheet showed

4) Sample preparation:

Species: mouse

Type of sample: primary cultured hippocampal neuron from mouse E18 embryonic

Sample preparation

The cultured hippocampal neuron was washed twice with 1XPBS, and then fixed with 4%PFA 14min or fixed with pre-cooled pure methanol 10min. Aften then cells were penetrated with 0.2% triton-X100 in 1XPBS for 8 min and blocked in 1% BSA in 1XPBS for 1 hour. Next cells were incubate with diluted antibody solution(1:500, as the product sheet say) at 4 degree overnight. The second day the cells were wash with 1XPBS for four times 5min each time. Then incubate with secondary antibody(Alexa 633 or Alexa 555 from Invitrogen 1:2000) for 1 hours at room temperature. After wash, finally the coverslip were mounted with prolong mounting solution(Invitrogen). The picture was taken with OMX superesolution microscope or zeiss fluorescent microscope.

Positive control: phalloindin 488 stain and or belta tubulin antibody stain.

Answer

Thank you for your reply.

Your protocol looks good. The one recommendation I could make would be to increase the dilution of the primary and secondary (especially) to see if that improves the staining.
You can try the primary at 1:250 and the secondary at 1:200 and see if that helps.

If this does not improve the staining, we would be happy to replace or refund the product. Please let me know if this does not help and what you would prefer as compensation.
