The secondary antibody I used was part of the DAKO ENVISION System. This system is based on an HRP labelled polymer that is conjugated to secondary antibodies. It can be used with both mouse and rabbit primary antibodies. Secondary antibody as well as the blocking reagent come ready to use so they don't need to be diluted. The blocking reagent for this kit is a peroxidase blocking step. It does not require you to use a protein blocking such as normal serum. You can do an extra normal serum blocking step if you have high background. For antigen retrieval, I incubated slides in citrate buffer and heated them for 20 minutes.
