      Luciferase from the firefly has become one of the more widely used reporter proteins for the study of gene expression. Luciferase catalyzes a bioluminescent reaction which requires the substrate luciferin as well as Mg2+ and ATP. Mixing these reagents with the cell extract containing luciferase, results in a flash of light that decays rapidly. This light can be detected by a luminometer. The total light emission is proportional to the luciferase activity of the sample.

      Hi this is how we detect Luciferase by other method.

      Here is an example of the IVIS imaging of these mice. Lysates blotted were generated from the paw tissue of these mice.

      Please can we try alternative antibody?

      Thank you for your email and providing the required data.

      I could replace the antibody at no cost however as we do not have received complaints about this lot so I am wondering the problem might be either with the expression of luciferase or with the dilution of the antibody. The 1/5000 dilution recommended on the datasheet is for guidelines only so 1/2000 dilution or loading high concentration of protein might be tried for optimization.

      The provided data does not show which cell type was used for lysates preparation so I would like to ask if you have confirmed the expression of luciferase by other methods. if yes, could you share the details?

      I am afraid the antibody anti-firefly luciferase antibody ordered for western blotting - AB81823. PO 355196786, is not working.

      We have tested the antibody a couple of times now on whole cell lysates from our Lucifierase transgenic mice.

      AbCam states in there guarantee that they will "Replace or refund for products not performing as stated on the datasheet".

      We would like to have replacement for this antibody with another that Abcam also state, and show, works in WB - AB99944.

      Please can you confirm we can try AB99944 as replacement?

      So basically i want to to do is pull-downs with Mouse monoclonal [Luci17] and do a western with anti-firefly raised either in goat or rabbit (if possible with HRP for ECL but not required).
