Additional Notes
Liver medial lobe was dissected, snap frozen in dry ice + isopropanol, and cryosectioned at 8um thickness
Antigen retrieval was performed by incubation in 10 mM sodium citrate buffer, pH 6, in rice cooker for 20 minutes
In the image, blue = Hoechst 33342 nuclear stain and green = anti-F4/80. A control in which secondary but not primary Ab was added is provided for reference.
Unfortunately, we observed no clear patterns that would indicate successful staining, and the observed colors are likely due to autofluorescence. Therefore, our findings are inconclusive.
Abcam response
Thank you for your valuable feedback. IHC-Fr is not a tested application for ab6640 and it seems that this antibody does not recognize F4/80 under these experimental conditions. Thank you for your time - we hope that this information will be helpful to other researchers.
