    Pseudomonas aeruginosa produces two extracellular protein toxins, Exoenzyme S and Exotoxin A. Exoenzyme S is probably an exotoxin. It has the characteristic subunit structure of the A-component of a bacterial toxin, and it has ADP-ribosylating activity (for a variety of eukaryotic proteins) characteristic of exotoxins. It has been suggested that exoenzyme S may act to impair the function of phagocytic cells in the bloodstream and internal organs to prepare for invasion by Ps. aeruginosa. Exoenzyme S is a potent stimulus for the proliferation of T cells in greater than 95% of adults, which could contribute to the pathogenesis of Cystic Fibrosis.

I am have problems binding the antibody ab20031 to magnetic dynabeads despite meticulously following the recommended protocol and having tried a wide variety of buffers. The spec sheet for the beads insists that the coupling buffer must not contain certain reactive groups as this interferew with coupling efficiency. Can you therefore please confirm that the buffer the antibody is provided in does not contain the following groups - amines, thiols or hydroxyls (e.g. as found in tris, glycine or proteins other than the desired antibody)? I would like to confirm that ab20031 is supplied in PSB + sodium azide at 0,02 %. Please, note that this antibody was raised in hens and that the IgY molecule has slightly different characteristics than rabbit IgG. I would recommend checking the coupling protocol for the beads you are using to assure that also IgY can be coupled to those beads. Please also note that the product is a total IgY, not affinity purified and its purity is 80 % (measured by a total IgY fraction estimatation). There can therefore be other contaminating proteins present in this solution, but their characteristics are not known to us.

I just tried to contact Abcam to ask some technical questions about the product that I just purchased. I bought the Chicken Polyclonal to Exoenzyme S (ab20031) for the western blot study of Pseudomonas aeruginosa isolates. I'm just not sure that which strian of P aeruginosa that you used as a positive control. Did you test on the laboratory standard PAO strain or other strains else? Here I have only PAO as a reference, but from one paper said that it produces low level of Exoenzme S (Grimwood 1989 Antimicrobial agents and chemotherapy). I appreciate any suggestion from you and look forward to hearing from you.

Chicken polyclonal to Exoenzyme S (ab20031) was raised against the C-terminal part of Exoenzyme S, which is conserved in many different Pseudomonas aeruginosa strains, and therefore this antibody should recognize Exoenzyme S from many strains. In this specific case, the sequence of Exoenzyme S from the Pseudomonas aeruginosa strain PAO1 was used.
