RNA 修飾の概要
エピジェネティクスの研究には、多くの新しくエキサイティングな分野がありますが、その一つに RNA 修飾(RNA modification)があります。RNA 修飾の検出法やシーケンシング法の発展から miCLIP などのような新しい技術が開発されました。それによって近年新たな RNA 修飾が、さまざまな細胞あるいは生物種で発見され、またマッピングされるようになっています。現在存在が明らかになっている RNA 修飾には N6-methyladenosine(m6A) の他、m1A、m2,2G など、100 以上あります。これらの修飾は mRNA、tRNA、rRNA の他、miRNA(micro RNA)などの non-coding RNA においても存在し、各修飾それぞれが RNA の構造、輸送、安定性、スプライシングなど、独自の機能を有することが明らかになっています。しかしながら機能や分布が明らかになっていない修飾の方が多く、この分野は未知の魅力に富んでいると言えます。
mRNA 中の RNA 修飾
RNA 修飾は mRNA にも存在し、RNA 構造の変化、RNA-タンパク質結合の促進、RNA の電荷の変化、塩基対の形成など、mRNA の機能に必須なものもあります。中でも m6A は mRNA 内で最も豊富に存在する修飾の一つであり、mRNA のスプライシング2、構造変化3、安定性4、翻訳効率5など、多数の機能に関与していることが知られています。m6A の機能と分布につきましてはこちらもご参照ください。
tRNA 中の RNA 修飾
RNA の中で最も多くの RNA 修飾が存在するのは tRNA であり、RNA 修飾は5 塩基に 1 つ程度あると考えられています6。また tRNA 中の修飾はその種類が多様であり、またその形成のために多種の酵素が他段階に反応する必要があるような修飾も存在します7。tRNA 中の RNA 修飾の多くはアンチコドン・ループ(anticodon loop)に存在し、その機能はコドンとアンチコドンの相互作用をサポートし、フレームシフトを防ぎ、翻訳効率を高めることであると考えられています8。
