Protein kinases are enzymes that transfer a phosphate group from a phosphate donor, generally the g phosphate of ATP, onto an acceptor amino acid in a substrate protein. By this basic mechanism, protein kinases mediate most of the signal transduction in eukaryotic cells, regulating cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement and cell movement, apoptosis, and differentiation. With more than 500 gene products, the protein kinase family is one of the largest families of proteins in eukaryotes. The family has been classified in 8 major groups based on sequence comparison of their tyrosine (PTK) or serine/threonine (STK) kinase catalytic domains. The tyrosine kinase (TK) group is mainly involved in the regulation of cell-cell interactions such as differentiation, adhesion, motility and death. There are currently about 90 TK genes sequenced, 58 are of receptor protein TK (e.g. EGFR, EPH, FGFR, PDGFR, TRK, and VEGFR families), and 32 of cytosolic TK (e.g. ABL, FAK, JAK, and SRC families). -

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Eph receptor A4/SEK antibody (ab5389)
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human breast carcinoma (BC) tissue labeling Eph receptor A4/SEK with ab5389 at 1/25. Tissue was fixed with formaldehyde and blocked with 3% BSA for 0.5 hour at 37°C. Heat mediated antigen retrieval was performed using a citrate buffer (pH6). Samples were incubated with primary antibody (1/25) for 1 hour at 37°C. A Peroxidase-conjugated Goat anti-rabbit polyclonal (ready to use) was used as the secondary antibody.

Flow cytometry analysis of CEM cells labelling Eph receptor A4/SEK (green) with ab5389 compared to a negative control (blue). The cells were fixed with paraformaldehyde and then permeabilized with 90% methanol for 10 min. The cells were then incubated in 3% BSA to block non-specific protein-protein interactions followed by the antibody (ab5389, 1µg/1x106 cells) for 60 min at 37ºC. The secondary antibody used was FITC conjugated Goat anti-Rabbit antibodies at 1/200 dilution for 40 min at room temperature. Blank control (blue line) was used under the same conditions. Acquisition of >10,000 events was performed.
