Question

We have tried many ways, like different Ab dilution, wash times, sample preparation et al. The result is not good, as you can see the Attached typical gel image. I also showed the images to Jason when Abcam had a show in UGA this week. Can you send us an alternative Dusp2/7 antibodies ?

I just wanted to check in with you regarding the issues that you had been experiencing the DUSP7 and DUSP2 antibodies.

Have you had an opportunity to assemble information about your protocol? We take issues such as these seriously and we guarantee our products for a period of 6 months if used the manner specified on our datasheets. We ask for protocol details to determine if the product is being used consistent with the Abpromise.

It is also very important that we have the Abcam confirmation number or the PO number used to purchase the product as I would need these to issue a replacement or refund.

I am sorry about the issues you have been having. Thank you for bringing this to our attention here and with Jason. Unfortunately the image did not arrive with this. It may be that the file size is too large for our email system.

As company policy, in order to send a replacement we do need the particulars of your protocol. This information is important not only in helping us understand the problems that you are experiencing but in our internal testing as well.

I see that you have been working with Mouse ESCs, could you tell me the lysis buffer that you have used?
Have you reduced and denatured your samples? Which method/buffers did you use?
How much total protein did you use per well?
Did you load a loading control? Which lc did you use? What were the results?
Did you load a positive control? Which pc did you use? What were the results?
What membrane type did you transfer to?
What blocking steps did you use?
What dilutions of this product did you use? How long and at what temperature was this antibody incubated?
What was the secondary antibody that you have used?How long and at what temperature was this antibody incubated?
Have you run a no primary antibody control blot? What were the results?
What is the wash buffer that you use?
Could you provide the Abcam order confirmation number or PO number used to purchase this?

I am writing about Abs questions from Abcam.

Abcam provides good antibodies, and we are happy to order products from Abcam. We ordered Dusp2 (ab95159) and Dusp7 (ab100921) from Abcam, and used them for WB in mouse embryonic stem cells. But we found the antibody is not good as showed in the websites. There were a lot of unspecific bands (we can send you images later), and the predicted bands are very weak compared to other unspecific bands.

So I wonder if we can replace them or get the money back?

I am sorry to hear of the difficulties that you have been experiencing using these products. Often times high background can be reduced by using TBS + 0.1% Tween 20 for your washes or by reducing the concentration of your antibodies. Would you mind sending me the details of your protocol including the tissue type you are working with, your sample preparations including denaturing and reducing steps, the amount of total protein per well, the positive controls you have used, loading controls or poncheau staining results, blocking procedures, antibody incubation steps, washes, images and any steps that you may have changed while troubleshooting.

These details will not only help me to better understand your difficulties and be able to offer suggestions but will also be used for internal testing of the products if deemed neccessary. If we are unable to solve the problems you are experiencing we will replace or refund the antibody according to our Abpromise guarantee.
