Defects in DSG2 are the cause of familial arrhythmogenic right ventricular dysplasia type 10 (ARVD10) [MIM:610193]; also known as arrhythmogenic right ventricular cardiomyopathy 10 (ARVC10). ARVD is an autosomal dominant disease characterized by partial degeneration of the myocardium of the right ventricle, electrical instability, and sudden death. It is clinically defined by electrocardiographic and angiographic criteria; pathologic findings, replacement of ventricular myocardium with fatty and fibrous elements, preferentially involve the right ventricular free wall.
Genetic variations in DSG2 are the cause of susceptibility to cardiomyopathy dilated type 1BB (CMD1BB) [MIM:612877]. A disorder characterized by ventricular dilation and impaired systolic function, resulting in congestive heart failure and arrhythmia. Patients are at risk of premature death. -

Immunocytochemistry/ Immunofluorescence - Anti-Desmoglein 2/DSG2 antibody [6D8] - BSA and Azide free (ab269978)
This data was developed using the same antibody clone in a different buffer formulation containing PBS and sodium azide (ab14415).
ab14415 staining Desmoglein 2 in A431 cells. The cells were fixed with 4% PFA (10min), permeabilized with 0.1%PBS-Tween for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1%PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab14415 at 5μg/ml and ab6046, Rabbit polyclonal to beta Tubulin - Loading Control, at 1/1000 dilution. Cells were then incubated with ab150117, Goat Anti-Mouse IgG H&L (Alexa Fluor® 488) at 1/1000 dilution (shown in green) and ab150084, Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (Alexa Fluor® 594) at 1/1000 dilution (shown in pseudocolor red). Nuclear DNA was labeled with DAPI (shown in blue).
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Performed under reducing conditions.This data was developed using the same antibody clone in a different buffer formulation containing PBS and sodium azide (ab14415).
ab14415 was shown to specifically react with DSG2 (Desmoglein 2) in wild type HAP1 cells. No band was observed when DSG2 (Desmoglein 2) knockout samples were used. Wild-type and DSG2 (Desmoglein 2) knockout samples were subjected to SDS-PAGE. The membrane was blocked for an hour using 5% milk before ab14415 and ab181602 (Rabbit anti-GAPDH loading control) were incubated overnight at 4°C at 3ug/ml and a 1/20000 dilution respectively. Blots were developed with Goat anti-Rabbit IgG H&L (IRDye® 800CW) preadsorbed (ab216773) and Goat anti-Mouse IgG H&L (IRDye® 680RD) preadsorbed (ab216776) secondary antibodies at 1/20000 dilution for 1 hour at room temperature before imaging.
