    Thanks for your email. I look forward to information about a new lot of
    anti-CXXC5 if this becomes available in the future? Please note that the
    current lot does recognise CXXC5 protein, but that in my hands this
    detection is extremely weak compared to the detection of the ˜120 kDA
    band(s)
    Im not quite sure why you are suggesting I try an antibody against P2Y6
    protein and how this would help my current research?? So I would decline
    this offer (unless Im misunderstanding something, or that you have made a
    misprint and were referring to something else?),

    Firstly, have you BLAST searched the peptide sequence used as the CXXC5
    immunogen against the human protein database?? If you use a low stringency
    search you might have matches to other proteins...... and are any of these
    at ˜120 kDa?? The antibody is predominantly recognising this species and
    works well on western and IP for this species. It seems a waste not to try
    to ID it if it could be done simply.
    Secondly, are you offering a free replacement CXXC5 antibody with a
    different lot number?? In case that particular bleed from the rabbit has a
    different profile?? If that is the case, then yes I would be happy to try
    this.

    We unfortunately do not have new lot for this product at the moment however lab has agreed to retest the lot; I will share the results with you soon.

    Thank you very much for having patience!

    I have requested the exact immunogen information from our collaborator and is now waiting a reply from them. I will send you the details soon.

    Currently, we do not have explanation for 120 kDa band; this could be due to non specificity of antibody or due to lysates. The antibody however should detect a 30-35 kDa band.

    The idea for replacement was to try a different lot and then seeing, if the new lot gives same result; if yes, then it will be worth identifying the 120kDa band.

    We do not have new lot instock however I have requested the lab for its avialability.

    Thank you very much for providing the protocol details and images.

    The protocol seems fine to me however the following protocol optimizations could;

    - At Abcam we use 5% BSA for blocking; I would recommend trying BSA instead of milk.
    - Boil the lysates for 10 minutes at 100C.

    We are not sure about the identity of 120 kDa band so I am sorry we will not be able to comment on this. The immunogen of this antibody is from the N-terminal region of human CXXC5 (http://www.uniprot.org/uniprot/Q7LFL8; this database actually shows 2 isoforms of CXXC5).

    I can offer you a different replacement antibody if you would like to compare the results. Could you let me know how you would like to proceed.

    Inquiry: Our lab purchased this for analysis of CXXC5. Our experience with the western blots is that there is a major species at around ˜120 kDa, a number of other cross reactive bands and that a protein of the predicted size for cxxc5 ˜35 kDa, was only a very minor species on the western and only really seen on a very long exposure (I have attached a western for your interest).....in this respect, I noted with interest that the image on the datasheet for this antibody has been cut at above ˜45 kDa. We are interested in pursuing Cxxc5 and so we have also used the antibody in immunoprecipitation. Again the major species immunoprecipitated is at ˜120 kDa. By siRNA knockdown, we have the impression that the very weak immunoreactive band at the predicted size may indeed be cxxc5...but the performance of the antibody(sold as an anti-CXXC5) is very poor. And our evidence indicates that it is primarily recognising a 120 kDa species. Would it be possible to know the sequence of the peptide immunogen so that we could try and search the protein databases as to the identity of the 120 kDa band? Or could you do this and tell us what you think the 120 kDa protein might be??? Can we exchange/get our money back for this poor anti-cxxc5 reagent ?
