Immunocytochemistry/Immunofluorescence analysis of Jurkat (Human T cell leukemia cell line from peripheral blood) labelling CD99 with purified ab108297 at 1/500. Cells were fixed with 4% PFA and permeabilized with 0.1% triton X-100. ab150077 Goat anti rabbit IgG (Alexa Fluor^® 488) at 1/1000 was used as the secondary antibody. Nuclei were counterstained with DAPI. PBS was used instead of the primary antibody as the negative control.

CD99 was immunoprecipitated from 0.35 mg HUVEC (Human umbilical vein endothelial cell) whole cell lysate 10 µg with 108297 at 1/50 dilution (2µg). VeriBlot for IP Detection Reagent (HRP)(ab131366) was used at 1/5000 dilution.

Lane 2: ab108297 IP in HUVEC whole cell lysate

Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab108297 in HUVEC whole cell lysate

Thank you for your enquiry and your interest in our products.

Each antibody should be titrated to determine the optimal working dilutions. It is the customer's responsibility to calculate how much product is needed for each assay.

The required quantity of the antibody depends on many factors:

1. dilution factor of the antibody,

2. expression levels of the target,

3. how you carry out the assay (in duplicates, triplicates or in quadruplicates),

4. presence of positive and negative control,

5. surface area of the tissue section on the slides etc.

Question

I am looking for primary antibody against mks-5 for work with human cells. Application is immunofluorescence microscopy. I was wondering if you had suggestions of products that would fit my requirements.

Thank you for contacting Abcam.

We currently do not have any product against the C.elegans protein MKS-5 nor will our products against a similar proteinRPGRIP1L recognize MKS-5. A protein with a similar name is MSK5X (also known as CD99). We do have a large selection of antibodies against this target. Ab8855 (https://www.abcam.com/CD99-antibody-12E7-ab8855.html) has been tested in ICC/IF while ab108297 (https://www.abcam.com/CD99-antibody-EPR3096-ab108297.html) and ab75858 (https://www.abcam.com/CD99-antibody-EPR3097Y-ab75858.html) are both guaranteed to work in ICH and ICC on human samples.

Abcam also carries a number of reagents for IF staining, including serums and blocking reagents, mounting media and background reducing buffers. OurBrightMount Plus Aqueous Mounting Medium for Fluorescent Staining (ab103748;https://www.abcam.com/BrightMount-Plus-25ml-Aqueous-Mounting-Medium-Anti-fading-for-Fluorescent-Staining-ab103748.html) is specifically designed for use in IF and would protect against photo-bleaching of your samples.
