    • 機能

      Required for the function of light chain amino-acid transporters. Involved in sodium-independent, high-affinity transport of large neutral amino acids such as phenylalanine, tyrosine, leucine, arginine and tryptophan. Involved in guiding and targeting of LAT1 and LAT2 to the plasma membrane. When associated with SLC7A6 or SLC7A7 acts as an arginine/glutamine exchanger, following an antiport mechanism for amino acid transport, influencing arginine release in exchange for extracellular amino acids. Plays a role in nitric oxide synthesis in human umbilical vein endothelial cells (HUVECs) via transport of L-arginine. Required for normal and neoplastic cell growth. When associated with SLC7A5/LAT1, is also involved in the transport of L-DOPA across the blood-brain barrier, and that of thyroid hormones triiodothyronine (T3) and thyroxine (T4) across the cell membrane in tissues such as placenta. Involved in the uptake of methylmercury (MeHg) when administered as the L-cysteine or D,L-homocysteine complexes, and hence plays a role in metal ion homeostasis and toxicity. When associated with SLC7A5 or SLC7A8, involved in the cellular activity of small molecular weight nitrosothiols, via the stereoselective transport of L-nitrosocysteine (L-CNSO) across the transmembrane. Together with ICAM1, regulates the transport activity LAT2 in polarized intestinal cells, by generating and delivering intracellular signals. When associated with SLC7A5, plays an important role in transporting L-leucine from the circulating blood to the retina across the inner blood-retinal barrier.

During gestation, expression in the placenta was significantly stronger at full-term than at the mid-trimester stage. Expressed in HUVECS and at low levels in resting peripheral blood T-lymphocytes and quiescent fibroblasts. Also expressed in fetal liver and in the astrocytic process of primary astrocytic gliomas. Expressed in retinal endothelial cells and in the intestinal epithelial cell line C2BBe1.

      Question

      Inquiry: The CD98 protein is a heterodimer, composed of a heavy chain and a light chain linked by disulfide bonds. We're looking for an antibody staining ONLY the HEAVY CHAIN of CD98 (also named CD98hc, 4F2hc or SLC3A2) by immunofluorescence. The antibody clone MEM-108 is described as recognizing CD98, but it is not clear whether it would stains one or the other (or both) of the chains. The western-blot shown in the datasheet does not give a clear answer because the high molecular weigth signal suggests that the western-blot was done on a native gel (the expected size of the heterodimer is 125kDa whereas the heavy chain is approx. 80kDa and the light-chain 40kDa). Even if the heavy chain is highly glycosylated, we should expect a smaller band than what is shown on the figure... Do you have more info about the specificity of this antibody or about how this western-blot was performed ? Many thanks in advance for your help.

      ab23495 [44D7] reacts with the heavy chain of the CD98 molecule. Tested in ICC/IF.
      ab108300 [EPR3548(2)] recognises the extracellular region of the human CD98 (heavy chain).

      Thank you for contacting us with your question. We previously carried the non-conjugated CD98 antibody from the clone 4F2 as ab33232. The clone number for this antibody was changed by the production lab, and we now carry this antibody as ab23495 with the clone name "44D7". I can assure you that ab23495 is the same antibody as ab33232, and the clones 44D7 and 4F2 are the same clones. I hope that this information is helpful, but please let me know if you have any questions about this. I apologize for any confusion about the clone names.
