    Abcam's Caspase Family Colorimetric Substrate Kit Plus provides ready-to-use colorimetric substrates for members of caspase family proteases. All substrates are provided in liquid ready-to-use form.


    This kit is the idea tool to test which caspases are activated when apoptosis is induced in a particular cell type.

    This product is manufactured by BioVision, an Abcam company and was previously called K136 Caspase Colorimetric Substrate Set Plus. K136-7-25 is the same size as the 25 test size of ab102486.

    Abcam has not and does not intend to apply for the REACH Authorisation of customers’ uses of products that contain European Authorisation list (Annex XIV) substances.

    Caspases are members of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce 2 subunits, large and small, that dimerize to form the active enzyme.

Question

As I am testing the anticancer property of my compound against the cancer cells, I wish to check the caspase activity of all caspases from 1- 9 . I really appreciate If u can give me some details of any set of panel of caspases from 1- 9 for 1- 5 assays, since I dont want to buy it separately for each one. I am looking forward to hear from you soon.

We do have some panels for caspase activity measurement:

1) Ab102486: Includes substrates for caspases 1,2,3,5,6,8, and 9. It does not include substrates for caspases 4 and 7. There are enough reagents included to run 25 assays.

2) Ab102487: Includes substrates for caspases 1,2,3/7,4,5,6,8,9, and 10. Please note that caspases 3 and 7 will be measured together. There are enough reagents included to run 25 assays.

3) Ab102488: Includes substrates for caspases 1,2,3,4,5,6,8, and 9. It does not include substrate for caspases 7. There are enough reagents included to run 25 assays.

Would one of these kits work for you? It doesn't seem like they are exactly what you had in mind but they are the closest products that we currently carry in our catalog.

Gracias por tu paciencia.

Me han confirmado mis compañeros que este kit es compatible con cualquier muestra procedente de mamífero, incluyendo muestras de ratón.

El kit contiene diferentes sustratos con distintas sensibilidades, por lo que no nos es posible ofrecer un valor fijo de sensibilidad.

El kit solo se ha testado con células, pero según mis compañeros con tejido funcionará también, simplemente os aconsejamos homogeneizar unos 10mg de tejido en 100ul de buffer de lisis y después continuar a partir del apartado 3 del protocolo.
