    ab22687 recognises proenzyme (43 kDa) and the large subunit (19 kDa) and small (10 kDa) subunits of mature enzyme. For the mature enzyme, reactivity is strongest with the large subunit. No cross reactivity with other human caspases tested (1 to 3, 5 to 10, 14).

Optimal dilutions/concentrations should be determined by the end user.

    Thank you for contacting us and sorry for the delay in getting back to you.

    I can now share with you that as discussed over the phone,this antibody is supplied as whole rabbit serum and theconcentration has therefore not been determined.However, I can confirm that therecommended dilution for this antibody forWestern blottingis1:2000-1:4000. This has been amended fromthe original recommendation of 1/8000-1/16000.

    Last time you kindly advise our customer these suggestion below, our customer had change their condition as you recommended. 2) Load less sample protein i.e. ~20 ug/lane. 5) The recommended dilution in WB is 1/8000 - 1/16000 using Alkaline phosphatase-linked secondary antibody. Since you used an HRP-conjugated secondary in ECL+, which is generally more sensitive than the AP detection, I suggest diluting ab22687 at ~1:20000-300000 and incubating at room temp for 1 hr or 4C overnight. protein loading 20 ug blocking:4?, overnight 2nd Ab: dilute 1/200, 000(jackson innunoresearch) We would be appreciated if you could provide them the credit note or a replacement vial.
