        Flow Cytometry - Anti-C5a-R antibody [S5/1] (ab11867)

        Human peripheral blood granulocytes stained with ab11867 (red line). Human whole blood was processed using a modified protocol based on Chow et al, 2005 (PMID: 16080188). In brief, human whole blood was fixed in 4% formaldehyde (methanol-free) for 10 min at 22°C. Red blood cells were then lyzed by the addition of Triton X-100 (final concentration - 0.1%) for 15 min at 37°C. For experimentation, cells were treated with 50% methanol (-20°C) for 15 min at 4°C. Cells were then incubated with the antibody (ab11867, 1μg/1x106 cells) for 30 min at 4°C. The secondary antibody used was Alexa Fluor® 488 goat anti-mouse IgG (H&L) (ab150113) at 1/2000 dilution for 30 min at 4°C. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 1μg/1x106 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >30,000 total events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter. Gating strategy - peripheral blood granulocytes.

      I am seeing extra bands on my Western with this antibody. Do you have an image of a Western done with this antibody? If not, did you see only one band on the Western, or multiple bands?

      Thank you for your enquiry. Please note as far as we know this application is not described in an article yet. We received the information that this antibody is useful for Western blot from the researcher who provided us this antibody. They give us the information a single band was observed. Please note the observed molecular weight is 43-48 kDa on HL-60 cells and 50-55 kDa on eosinophils. So the molecular weight is depending on the type of cells. I hope this information helps.

      Thank you for your enquiry. For flow cytometry, the dilutions to be used depend on the detection system applied. It is recommended that users test the reagent and determine their own optimal dilutions. The typical starting working dilution is 1:50. The following article is a reference for the clone: Opperman, M et al; Probing the human receptor for C5a anaphylatoxin with side-directed antibodies. J Immunol 1993, 151: 3785 In this article they use 2ug/10E6 cells of FITC-labeled mAb. Please note that we are not able to provide FITC-labeled antibody. For inhibition of binding of C5a to its receptor dilutions have to be made according to the amounts of C5a to be inhibited.
