    Activation of C5 by a C5 convertase initiates the spontaneous assembly of the late complement components, C5-C9, into the membrane attack complex. C5b has a transient binding site for C6. The C5b-C6 complex is the foundation upon which the lytic complex is assembled.
    Derived from proteolytic degradation of complement C5, C5 anaphylatoxin is a mediator of local inflammatory process. It induces the contraction of smooth muscle, increases vascular permeability and causes histamine release from mast cells and basophilic leukocytes. C5a also stimulates the locomotion of polymorphonuclear leukocytes (chemokinesis) and direct their migration toward sites of inflammation (chemotaxis).

    Defects in C5 are the cause of complement component 5 deficiency (C5D) [MIM:609536]. A rare defect of the complement classical pathway associated with susceptibility to severe recurrent infections, predominantly by Neisseria gonorrhoeae or Neisseria meningitidis.
    Note=An association study of C5 haplotypes and genotypes in individuals with chronic hepatitis C virus infection shows that individuals homozygous for the C5_1 haplotype have a significantly higher stage of liver fibrosis than individuals carrying at least 1 other allele (PubMed:15995705).

    I was wondering if this product,Anti-C5 antibody (ab11898) mouse reactive, has even been tested in any functional neutralizing studies on mouse C5. Additionally, what are the purification steps taken on the polyclonal antibody prep before the final quantification and aliquoting of the supplied 50 ug?

    The C5 antibody ab11898 has not, to our knowledge, been tested in a neutralization assay.

    The antibody is purified over a protein G column to separate immunoglobulin from other proteins.

    I am very thankful for your kind help but in my second mail I have mentioned about complement proteins C5 antibody Anti-C5 antibody (ab11898) and may request is same as I asked about first one.
    What dilution is suitable for IHC in rat brain? because you have already mentioned 1/10 dilution forAnti-C5 antibody (ab11898) and you people provide 50ug, so if weconsiderthis volume than this is use full for only few reaction.

    Thank you very much for your reply, and I apologize that I did not mention the details of ab11898.

    This antibody has not been tested in our labs in IHC or for reactivity with the mouse protein, so we do not have a specific dilution or concentration to recommend for your assay. IHC was added to our datasheet due to feedback in literature showing that the antibody is suitable for this application, though I could not find a reference for the 1:10 recommended dilution. Since we are not sure whether the antibody will cross-react with rat samples, I would recommend using the ab33948 antibody that we discussed previoiusly.

    Thank you for your enquiry. For ab11898, the antibody should recognize the whole mouse C5 chain; since this is a polyclonal antibody it is very likely that it recognizes both C5a and C5b. As the sequence of the immunogen used is commercially sensitive, we can't guarantee this however. For ab17930 and ab17457, the antibodies only recognize non-reduced C5 where the molecule is not split into its component subunit chains. The antibodies are expected to recognize an epitope on the interface between the different subunits, though that is purely speculation. All three antibodies are £ 198.00.     Thank you for your questions regarding ab11898. This antibody does not cross react with human C5. The polyclonal antibody was generated with recombinant human protein from E.coli. In our product range we have the following antibodies which cross react with human C5: ab11876: monoclonal antibody to human C5/C5a, clone 557. ab11877: monoclonal antibody to human C5/C5a, N-terminus, clone 561. ab11879: monoclonal antibody to human C5/C5b, clone 568. As positive control liver tissues (hepatocytes) can be used as the primary site of synthesis is the liver. Secondary sites are lung, spleen, fetal intestine, monocytes, macrophages and type II alveolar cells. I hope this information will be helpful to you.
