    Clostridium botulinum neurotoxins are zinc-dependent proteases that block the release of acetylcholine from peripheral cholinergic nerve endings by targeting specific membrane proteins including synaptosomal-associated proteins (SNAP), vesicle-associated membrane proteins (VAMP), and syntaxins. Neurotoxin A is characteristically associated with human botulism and known to target and hydrolyse the 197-Gln-|-Arg-198 bond of SNAP-25.

What type of ELISA was done to validate these antibodies? How much protein was used and can we provide protocol details?

For catalog # ab20641 and ab20786 a direct ELISA was performed. The limit of detection (LOD) with direct ELISA is greater than 1:10,000 on both products with no amplification using OPD (o-phenylenediamine dihydrochloride) development and HRP conjugated 2nd antibody.
This is the most detail that we have available, and we recommend optimization of the reagents/antibodies in your specific application.

We previously ordered ab20641-250, and have product information describing this antibody as anti-"toxin." However, your website now calls this same product anti-"toxoid," has the product changed recently, or only the information describing the product? We are planning to repeat an experiment, and would like to be certain that we are using the same reagent. Any information you have regarding this situation would be much appreciated.

Thank you for your enquiry. I can confirm that the immunogen for this antibody (ab20641) has not changed. A toxoid is an attenuated/inactivated toxin. The Clostridium botulinum Type A Neurotoxin is inactivated using formaldehyde before it is used as an immunogen to create this antibody. Fairly recently, a decision was taken to change toxin to toxoid on the datasheet as this was considered to be a more accurate description. I hope this information is helpful.
