    Steroid hormone receptors are ligand-activated transcription factors that regulate eukaryotic gene expression and affect cellular proliferation and differentiation in target tissues. Transcription factor activity is modulated by bound coactivator and corepressor proteins. Transcription activation is down-regulated by NR0B2. Activated, but not phosphorylated, by HIPK3 and ZIPK/DAPK3.
    Isoform 3 and isoform 4 lack the C-terminal ligand-binding domain and may therefore constitutively activate the transcription of a specific set of genes independently of steroid hormones.

    Isoform 2 is mainly expressed in heart and skeletal muscle (PubMed:15634333). Isoform 3 is expressed by basal and stromal cells of prostate (at protein level) (PubMed:19244107).

    Sumoylated on Lys-388 (major) and Lys-521. Ubiquitinated. Deubiquitinated by USP26. 'Lys-6' and 'Lys-27'-linked polyubiquitination by RNF6 modulates AR transcriptional activity and specificity.
    Phosphorylated in prostate cancer cells in response to several growth factors including EGF. Phosphorylation is induced by c-Src kinase (CSK). Tyr-535 is one of the major phosphorylation sites and an increase in phosphorylation and Src kinase activity is associated with prostate cancer progression. Phosphorylation by TNK2 enhances the DNA-binding and transcriptional activity and may be responsible for androgen-independent progression of prostate cancer. Phosphorylation at Ser-83 by CDK9 regulates AR promoter selectivity and cell growth. Phosphorylation by PAK6 leads to AR-mediated transcription inhibition.
    Palmitoylated by ZDHHC7 and ZDHHC21. Palmitoylation is required for plasma membrane targeting and for rapid intracellular signaling via ERK and AKT kinases and cAMP generation.

    Question

    I have a big problem that I'm really hoping can be fixed. I wanted to order an anti-androgen receptor for my immunofluorescence studies but I accidentally ordered the mouse source (ab9474) and needed the rabbit source (ab3510). I have not used the antibody and will not be able to use it, as we are co-staining with another mouse antibody (cytokeratin, a cytoplasmic marker). Would it be possible for me to send the antibody back and switch it with the other or do you have any suggestions for my staining that may allow me to use both of the antibodies without cross-reactivity? Thanks so much!

    Answer

    There are two options for staining a tissue section with two mouse monoclonal antibodies. The first is to directly conjugate the antibodies, removing the need for any secondary antibodies. We have conjugation kits that go by the name "EasyLink". Here is a page from our website that lists them: www.abcam.com/easylink .

    The second approach is to use secondary antibodies that react exclusively with a single isotype subclass. This assumes that the two antibodies are different subclasses. Antibody ab9474 is IgG1. Do you know the subclass of the other antibody?

    Answer

    Thank you for your inquiry.

    We recommend to use this antibody at a dilution of 1/50in IHC-P. This is only a recommended starting dilution and may well have to be optimized by your customer according to their needs and samples.

    If the recommended dilution is used and ca 50ul per slide (this depends on the size of the sample on the slide, it needs to be covered all the way) then one vial of ab9474 will be enough for 250 slides.

    Answer

    Thank you for your interest. It is very likely that ab9474 works in mouse but we have no data yet. According to Blast search, the homology between human and mouse AR is very high. http://www.uniprot.org/blast/uniprot/2011101050RXMELAA8 Furthermore, we have some alternative antibodies against AR in our catalogue which proved to recognize mouse AR: ab2742, ab74272, ab3509, ab52615 Normal brain, epididymis, prostate, seminal vesicle; or skin and prostate cancer can be used as good positive control.
