Abcam’s Universal HIER antigen retrieval reagent (10X) is a heat-induced epitope retrieval (HIER) solution that is compatible with virtually all antibodies and eliminates the need for multiple buffers, including citrate buffer, EDTA or high pH Tris buffers. Antibody titers are doubled or tripled when compared to citrate buffer, pH 6.0. Universal HIER antigen retrieval reagent (10X) incorporates a color-coded high temperature pH indicator solution, allowing the end-user to check that the solution is at the correct dilution and pH by visual inspection. This product is specially formulated for superior pH stability at high temperatures in order to help prevent the possibility of losing pH-sensitive antigens. Universal HIER antigen retrieval reagent (10X) is non-toxic, non-flammable, odorless and sodium azide- and thimerosal-free.
ab208572 is the recommended HIER antigen retrieval reagent for PD-L1 RabMAb (clone 28-8, ab205921).

The Universal HIER antigen retrieval reagent is a buffered solution that is intended for laboratory use in heat induced antigen retrieval of formalin-fixed paraffin-embedded (FFPE) tissues for immunohistochemistry (IHC) procedures. The clinical interpretation of any staining or its absence should be complimented by morphological studies using proper controls and should be evaluated within the context of the patient's clinical history and other diagnostic tests by a qualified pathologist.

Immunohistochemistry of various tumor markers on brain sections. Slides were immersed in 1X universal antigen retrieval reagent (ab208572) and incubated for 20 min at 95 °C. Slides were stained to detect the expression of PRDX1 (ab41906), γ-H2A.X ( Anti-γ-H2A.X phosphorylated S139 antibody; ab2893), Ki 67 (ab 15580), 53BP1 (ab21083) and cleaved caspase 3 (ab13847) using an HRP/DAB detection method by using a mouse and rabbit specific HRP/DAB detection IHC kit (ab80436), following manufacturers recommendations with minor modifications.

Immunostaining for TNC within the blood vessel. Human lung samples were formalin-fixed and paraffin-embedded (FFPE). Deparaffinized and rehydrated 5-micron sections were subjected to antigen heat retrieval using citrate buffer (ab208572), for 30 min at 100 °C, cooled to room temperature for 20 min, treated with 3% hydrogen peroxide for 5 min, blocked for 1 h , and probed with primary antibody (TNC, 1:500, ab108930) overnight in 10% blocking solution.
