    DMAP1 is involved in transcription repression and activation. Its interaction with HDAC2 may provide a mechanism for histone deacetylation in heterochromatin following replication of DNA at late firing origins. Can also repress transcription independently of histone deacetylase activity. May specifically potentiate DAXX-mediated repression of glucocorticoid receptor-dependent transcription. Component of the NuA4 histone acetyltransferase (HAT) complex which is involved in transcriptional activation of select genes principally by acetylation of nucleosomal histone H4 and H2A. This modification may both alter nucleosome - DNA interactions and promote interaction of the modified histones with other proteins which positively regulate transcription. This complex may be required for the activation of transcriptional programs associated with oncogene and proto-oncogene mediated growth induction, tumor suppressor mediated growth arrest and replicative senescence, apoptosis, and DNA repair. NuA4 may also play a direct role in DNA repair when recruited to sites of DNA damage.

Answer

We have no clear explanation for the differences in reported mobility of DMAP detected utilizing ab3736, ab3737 and ab2848. The sequences of the peptides used fro ab3736 and ab3737 are proprietary. ab3736 it to a peptide that is toward the N-terminus of DMAP, but the peptide does not represent the extreme N-terminus of DMAP. ab3637 is to a peptide that is toward the C-terminus of DMAP, but the peptide does not represent the extreme C-terminus of DMAP. The peptide used for ab3637 is upstream of the peptide used for ab2848. Western blots using ab3636 and ab3637 have been run in parallel and recognize DMAP of the same relative mobility (60 to 65 kDa). Western blots using ab2848 have not been run in parallel with ab3636 and ab3637. Therefore, it is not known whether there is a difference in relative mobility of DMAP recognized by ab2848 vs ab3636/ab3637. Differences in observed mobility may be due to differences in the molecular weight standards and/or differences in running conditions. One possible reason is that different species samples migrate at a difference size as a result of motifications.
