Thank you for your honest and thoughtful feedback. I would recommend increasing the primary incubation time to overnight at 4C, as this is how Abcam tests all of its antibodies. Since this protein is also expressed in the cytoplasm, you may want to include a permeabilization step. Please let me know if these suggestions improve your results.
