    This is one of the several different receptors for 5-hydroxytryptamine (serotonin), a biogenic hormone that functions as a neurotransmitter, a hormone, and a mitogen. The activity of this receptor is mediated by G proteins that stimulate adenylate cyclase. It has a high affinity for tricyclic psychotropic drugs.

      IHC image of 5HT6 Receptor staining in Mouse Brain formalin fixed paraffin embedded tissue section, performed on a Leica Bond™ system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab103016, 0.5 µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

       

      For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

      5HT6 Receptor contains a potential glycosylation site (SwissProt) which may explain its migration at a higher molecular weight than predicted.

    J'ai testé en western-blot l'anticorpsAnti-5HT6 Receptor antibody (ab103016)

    Pour mon test, j'ai utilisé en contrôle négatif une lignée cellulaire n'exprimant pas le récepteur 5-HT6 de manière endogène (aucune réponse aux agonistes 5-HT6 en test AMPc), et en contrôle positif, cette même lignée transfectée avec le récepteur 5-HT6. Les dépôts correspondent à 10ug de protéines totales.

    J'obtiens bien une bande à la taille attendue, ainsi qu'une bande plus faible aux alentours de 40kD comme représenté sur la photographie de la notice, mais malheureusement, ces bandes sont aussi présentes dans mon contrôle négatif (voir fichier joint).

    J'ai utilisé cet anticorps dilué au 1/1000ème dans PBS/5%BSA/0,5%tween-20. Incubation sur la nuit à 4°C, 3 lavages de 10 minutes au PBS-Tween et ajout du secondaire anti-lapin couplé HRP (dilution 1/5000ème dans PBS-BSA-tween) 1h à température ambiante. La révélation correspond à une exposition de 5" dans un appareil chemidoc.

    Pourriez-vous me dire quel est votre contrôle négatif afin que je refasse un test ?

    Merci de nous avoir contactés et désolé pour le délai de notre réponse.
    J'ai contacté mes collègues du laboratoire, ils n'ont malheureusement pas testé cet anticorps avec un contrôle négatif. Les tests de caractérisation montrent une bande à ˜55 kDa avec des lysats de cerveau de souris.
    Afin de comparer et confirmer ou non les résultats obtenus avec le ab103016 je peux, si vous le désirez, vous envoyer une unité gratuite de ab101912, un autre anti-récepteur 5HT6 testé en Western Blot et chez la souris: www.abcam.com/ab101912 .
    Merci de me confirmer que vous acceptez cette résolution. Alternativement je peux également éditer un avoir contre la commande originale.
