Synthetic peptide conjugated to KLH derived from within residues 150 - 250 of Mouse Zfp57.
(Peptide available as ab45340.)
Our Abpromise guarantee covers the use of ab45341 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
|ChIP||Use at an assay dependent concentration. PubMed: 26728555|
|WB||Use a concentration of 1 µg/ml. Detects a band of approximately 57 kDa (predicted molecular weight: 48 kDa).Can be blocked with Mouse Zfp57 peptide (ab45340).|
Zfp57 KO ES cell lysates were kindly provided by our collaborator Prof. Anne Ferguson-Smith, University of Cambridge. The mutant mouse used to make these lysates is described here - Takahashi et al., Cold Spring Harb Symp Quant Biol. 2015;80:177-87.
This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using Licor blocking buffer before being incubated with ab45341 and anti-GAPDH as a loading control (shown in Red) overnight at 4°C. Antibody binding was detected using ab216773 at a 1:10,000 dilution for 1hr at room temperature and then imaged using the Licor Odyssey CLx.
Abcam recommends using milk as the blocking agent. Abcam welcomes customer feedback and would appreciate any comments regarding this product and the data presented above.
