WB Use at an assay dependent concentration. Detects a band of approximately 75 kDa (predicted molecular weight: 69 kDa).
IHC-P 1/50. Perform heat mediated antigen retrieval before commencing with IHC staining protocol. Boil tissue sections in 10 mM citrate buffer, pH 6.0 for 10 minutes followed by cooling at room temperature for 20 minutes.

    Predicted band size : 69 kDa
    Observed band size : 75 kDa (why is the actual band size different from the predicted?)
    Additional bands at : 46 kDa. We are unsure as to the identity of these extra bands.

    Exposure time : 12 minutes
    Sodium Iodide Symporter contains a number of potential glycosylation sites (SwissProt) which may explain its migration at a higher molecular weight than predicted.

Cells were fixed with 4% paraformaldehyde, permeabilized with 0.1% saponin in PBS and blocked with 5% BSA + 0.1% saponin in PBS for 1 hour at 37°C. Samples were incubated with primary antibody (1/50 in PBS + 1% BSA + 0.1% saponin) for 16 hours at 4°C. An Alexa Fluor® 488-conjugated Goat anti-mouse IgG polyclonal (1/300) was used as the secondary antibody.

  • ab17795 staining Sodium Iodide Symporter in Rat thyrocytes (Pccl3) by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with 4% paraformaldehyde, permeabilized with 0.1% saponin and blocked with 5% BSA for 1 hour at 37°C. Samples were incubated with primary antibody (1/50 in PBS + 1% BSA + 0.1% saponin) for 16 hours at 4°C. An Alexa Fluor® 488-conjugated Goat anti-mouse IgG polyclonal (1/300) was used as the secondary antibody.

  • Overlay histogram showing MCF7 cells stained with ab17795 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab17795, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in MCF7 cells fixed with 80% methanol (5 min)/permeabilized in 0.1% PBS-Tween used under the same conditions.
