Full length native protein (purified): MGSELETAME TLINVFHAHS GKEGDKYKLS KKELKELLQT ELSGFLDAQK DADAVDKVMK ELDENGDGEV DFQEYVVLVA ALTVACNNFF WENS, corresponding to amino acids 1-94 of Cow S100 (alpha chain). A mixture of alpha alpha homodimers and alpha beta heterodimers was used.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Use a concentration of 0.2 - 1 µg/ml.
Use a concentration of 1.67 - 10 µg/ml. Detects a band of approximately 11 kDa.
Use a concentration of 2.5 - 25 µg/ml.
Weakly binds calcium but binds zinc very tightly-distinct binding sites with different affinities exist for both ions on each monomer. Physiological concentrations of potassium ion antagonize the binding of both divalent cations, especially affecting high-affinity calcium-binding sites.
Highly prevalent in heart. Also found in lesser quantities in skeletal muscle and brain.
Belongs to the S-100 family. Contains 2 EF-hand domains.
Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-S100 antibody (ab34686)This image is courtesy of an anonymous Abreview
ab34686 staining S100 in Rat nerve tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 10% serum for 30 minutes at 25°C; antigen retrieval was by heat mediation in a 10mM citrate buffer, pH 6.0. Samples were incubated with primary antibody (1/250 in PBS + 1x casein) for 1 hour at 25°C. A Biotin-conjugated Goat anti-rabbit IgG polyclonal (1/200) was used as the secondary antibody.
Ab34686, at a 1/500 dilution, staining S100 in mouse brain tissue by immunohistochemistry. The tissue was formalin fixed and embedded in paraffin. Dark nuclear staining is observed within basket cells located near the Purkinje cells in the cerebellum.
