  • 関連性Platelet-activating factor (PAF) is a pro-inflammatory lipid mediator possessing a unique 1-O-alkyl-2-acetylsnglycero-3-phosphocholine backbone. The PAF receptor (PAFR), which belongs to the superfamily of seven transmembrane (7TM), G-protein coupled receptors (GPCRs), transduces pleiotropic functions including cell motility, smooth muscle contraction, and synthesisand release of mediators and cytokines via multiple heterotrimeric G-proteins. Pharmacological studies have suggested that PAF functions in a variety of settings including allergy, inflammation, neural functions, reproduction, and atherosclerosis. Guanine nucleotide-binding proteins (G proteins) are involved as a modulator or transducer in various transmembrane signaling systems. The G(i) proteins are involved in hormonal regulation of adenylate cyclase: they inhibit the cyclase in response to beta-adrenergic stimuli. The beta and gamma chains are required for the GTPase activity, for replacement of GDP by GTP, and for G protein-effector interaction
