関連性PTP lambda is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle and oncogenic transformation. PTP lambda is a large protein that possesses a C-terminus PTP domain and multiple noncatalytic domains which include a domain with similarity to band 4.1 superfamily of cytoskeletal-associated proteins, a region consisting of five PDZ domains and a leucine zipper motif. Four alternatively spliced transcript variants, which encode distinct proteins, have been reported.
PTP lambda is up-regulated upon cell contact and is found in high levels in brain, pancreas, and skeletal muscle; less in colon, kidney, liver, stomach and uterus. It is not expressed in placenta and spleen.

Paraffin embedded human cerebral cortex tissues were cut into 4µm sections and incubated with ab13223 (1/75) for 30 minutes at room temperature. Antigen retrieval was performed by heat induction in citrate buffer pH 6.ab13223 was tested in a tissue microarray (TMA) containing a wide range of normal and cancer tissues as well as a cell microarray consisting of a range of commonly used, well characterised human cell lines. ICC/IF image of ab13223 stained HeLa cells. The cells were 100% methanol fixed (5 min), permabilised in 0.1% PBS-Tween (20 min) and incubated with the antibody (ab13223, 5µg/ml) for 1h at room temperature. 1%BSA / 10% normal goat serum / 0.3M glycine was used to block non-specific protein-protein interactions. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red). DAPI was used to stain the cell nuclei (blue). This antibody also gave a positive IF result in 4% PFA and 100% methanol fixed Hek293, HepG2 and MCF7 cells. However, this Fast-Track antibody is not yet fully characterised. This image represents inconclusive preliminary data.
