Cross-links antiparrallel microtubules at an average distance of 35 nM. Essential for controlling the spatiotemporal formation of the midzone and successful cytokinesis. Required for KIF14 localization to the central spindle and midbody. -

Phosphorylation by CDK1 in early mitosis holds PRC1 in an inactive monomeric state, during the metaphase to anaphase transition, PRC1 is dephosphorylated, promoting interaction with KIF4A, which then translocates PRC1 along mitotic spindles to the plus ends of antiparallel interdigitating microtubules. Dephosphorylation also promotes MT-bundling activity by allowing dimerization. -

Immunocytochemistry analysis of PRC1 using ab119338 at 2µg/mL concentration shows staining in 4% paraformaldehyde-fixed 0.1% Triton X-100 permeabilized HeLa Cells. PRC1 (green), F-Actin staining with Alexa Fluor® 555 Rhodamine Phalloidin (red) and nuclei with SlowFade® Gold Antifade Mountant with DAPI (blue) is shown. Negative control has no primary antibody.

Immunofluorescent analysis of PRC1 using PRC1 Monoclonal Antibody (16F2) (ab119338) shows staining in U251 Cells. PRC1 (green), F-Actin staining with Phalloidin (red) and nuclei with DAPI (blue) is shown. Cells were grown on chamber slides and fixed with formaldehyde prior to staining. Cells were probed without (control) or with an antibody recognizing PRC1 (ab119338) at a dilution of 1:20 over night at 4oC, washed with PBS and incubated with a DyLight-488 conjugated secondary antibody. Images were taken at 60X magnification.
