ab75657 was affinity-purified from rabbit antiserum by affinity-chromatography using epitope-specific phosphopeptide. The antibody against non-phosphopeptide was removed by chromatography using non-phosphopeptide corresponding to the phosphorylation site.

Phosphorylation on Thr-500 within the activation loop renders it competent to autophosphorylate. Autophosphorylation on other sites i.e. in the N-terminal and hinge regions have no effect on enzyme activity.
