May play an important role in eliminating peroxides generated during metabolism. Might participate in the signaling cascades of growth factors and tumor necrosis factor-alpha by regulating the intracellular concentrations of H(2)O(2). Reduces an intramolecular disulfide bond in GDPD5 that gates the ability to GDPD5 to drive postmitotic motor neuron differentiation.

  • Predicted band size : 22 kDa

    Lane 1: Wild-type HAP1 cell lysate (20 µg)
    Lane 2: Peroxiredoxin knockout HAP1 cell lysate (20 µg)
    Lane 3: A431 cell lysate (20 µg)
    Lane 4: Jurkat cell lysate (20 µg)
    Lanes 1 - 4: Merged signal (red and green). Green - ab70666 observed at 23 kDa. Red - loading control, ab181602, observed at 37 kDa.
    ab70666 was shown to specifically react with Peroxiredoxin when Peroxiredoxin knockout samples were used. Wild-type and Peroxiredoxin knockout samples were subjected to SDS-PAGE. ab70666 and ab181602 (loading control to GAPDH) were diluted 1/20 000 and 1/10000 respectively and incubated overnight at 4°C. Blots were developed with Goat anti-Mouse IgG H&L (IRDye® 800CW) preadsorbed ab216772 and Goat Anti-Rabbit IgG H&L (IRDye® 680RD) preadsorbed ab216777 secondary antibodies at 1/10000 dilution for 1 h at room temperature before imaging.

    Incubated with the primary antibody overnight at 4°C. Incubated with the secondary antibody for 1 hour at room temperature.

  • Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human thyroid tissue labelling Peroxiredoxin 1 with ab70666.

  • Immunoprecipitation analysis using ab70666.

    Lane 1: HeLa cell lysate
    Lane 2: Immunoprecipitate from HeLa cell lysate (200 µg) using ab70666 (1 µg)
    Lane 3: Immunoprecipitate from HeLa cell lysate (200 µg) using ab70666 (2 µg)
    Lane 4: Immunoprecipitate from PBS using ab70666 (2 µg)

    ab70666 was also used for western blot at 1/20000 overnight at 4°C.

    Incubated with the secondary antibody at 1/5000 for 1 hour at room temperature.
