  • ab4666 staining Peripherin in pig small intestine tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 2% BSA for 10 minutes at 21°C; antigen retrieval was by heat mediation in citric acid. Samples were incubated with primary antibody (1/2500 in TBS/BSA/azide) for 2 hours at 21°C. A Biotin-conjugated Goat anti-rabbit IgG polyclonal (1/300) was used as the secondary antibody.

  • ab51424 staining Peripherin in goat small intestine sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 2% serum for 10 minutes at 21°C; antigen retrieval was by heat mediation in citric acid. Samples were incubated with primary antibody (1/1500 in TBS/BSA/azide) for 2 hours at 21°C. A Biotin-conjugated goat anti-rabbit IgG polyclonal (1/300) was used as the secondary antibody.

  • ab4666 at a 1/3000 dilution staining Peripherin in human small intestine tissue sections by Immunohistochemistry (paraffin embedded) incubated for 20 minutes at 25°C. Fixed in formaldehyde. Heat mediated antigen retrieval step performed using EDTA pH 8.0, for 20 minutes at 100°C. Secondary used undiluted polyclonal Goat anti-mouse/rabbit IgG.
    Note nerve fibers and ganglion cells in the submucosa, and between the circular and longitudinal muscle layers

  • ab4666 stained SKNSH cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody ab4666 used at 1/200 dilution overnight at +4°C. The secondary antibody (green) was a goat anti-rabbit DyLight® 488 (ab96899) IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

    Observed band size : 57 kDa (why is the actual band size different from the predicted?)
    Additional bands at : 48 kDa. We are unsure as to the identity of these extra bands.
