Fig. 1 shows the enrichment at promoter region compared to IgG ChIP.
Fig. 2 shows the loss of p400 at promoter and enhnancer region of gene upon PMA treatment. Since this protein is involved in exchange of H2A.Z , its lost from regulatory region upon gene induction.
Undilluted 5 ul antibody was used for each IP. The ChIP-IT kit from Active motif is used conduct IP and ChIP.
The cell from 15 cm plate are scraped after crosslinking, lysed and enzymatically digested as per protocol (Active motif) in 1 ml lysis buffer. For each IP 100 ul of chromatin is used.
