The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Before use, centrifuge ab123472 suspension at 10 000g for 10 min at 4°C. Discard the supernatant and resuspend the pellet into the desired buffer. Desalt the sample using either Sephadex G-25 or dialysis. Abs/Em = 651/662 nm.
The extinction coefficient of CL-APC at 650nm is 70,000 cm-1M-1.

Allophycocyanin alpha chain
Allophycocyanin beta chain
関連性Allophycocyanin is a protein from the light-harvesting phycobiliprotein family, along with phycocyanin, phycoerythrin and phycoerythrocyanin. It is an accessory pigment to chlorophyll. All phycobiliproteins are water soluble and therefore cannot exist within the membrane like carotenoids, but aggregate to form clusters that adhere to the membrane called phycobilisomes. Allophycocyanin absorbs and emits red light (650 & 660 nm max, respectively), is readily found in Cyanobacteria and red algae, and is a useful immunoassay tool.
APC is made up of alpha and beta subunits and is present as a trimer (alpha beta)3. The trimer is unstable and susceptible to dissociation at low concentrations. The monomer, alpha beta, has a lower fluorescence quantum yield compared to the trimer and the maximal absorption is shifted to 620 nm. The chemically crosslinked APC (CL-APC) trimer is much more stable than the native protein, but still retains comparable spectroscopic properties. The extinction coefficient of CL-APC at 650 nm is 700,000
