Abcam’s Human IL-1RA ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IL-1RA in cell lysates and tissue lysates.
This assay employs an antibody specific for Human IL-1RA coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-1RA present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-1RA antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-1RA bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

IL-1Ra measured in RIPA lysates from control cells (-) or cells stimulated for 24 hours with 50 ng x mL-1 of PMA (ab120297) with the addition of 1 ug x mL-1 of LPS (Sigma) (P/L) for the last 6 hours (IL-1Ra per million cells; duplicates +/- SD). 5e6 cells were lysed in 0.1 mL, and tested in the dilution range of 1/1-1/40.
IL-1Ra measured in culture supernatants from control (-) or activated PBMCs ((PHA) 48 hours with 2% PHA-M; LifeTechnologies) and in human biological fluids. Signals from plasma and serum were below level of detection (duplicates +/- SD).
