Recommended dilutions ChIP: Use 2 - 5 µg for 25 µg of chromatin ICC/IF: Use at a concentration of 1 µg/ml. WB: Use at a concentration of 1 µg/ml. Detects a band of approximately 17 kDa (predicted molecular weight: 15 kDa). Can be blocked with Histone H3 peptide - tri methyl K9 (ab1773). ELISA: Use at an assay dependent dilution.
Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

Histones are basic nuclear proteins that, together with DNA, make up the nucleosome structure in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer, which is composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin structure is further compacted through the interaction of a linker histone H1.
Methylation of specific residues in the histone tails is a central modification for regulating epigenetic transitions in chromatin. Modified lysine residues can exist in a mono-, di-, or tri-methylated state, whilst arginine residues can be mono- or di-methylated.

Chromatin was prepared from Hela cells according to the Abcam X-ChIP protocol. Cells were fixed with formaldehyde for 10 min. The ChIP was performed with 25 µg of chromatin, 2 µg of ab71604 (blue), and 20 µl of Protein A/G sepharose beads. No antibody was added to the beads control (yellow). The immunoprecipitated DNA was quantified by real time PCR (Taqman approach for active and inactive loci, Sybr green approach for heterochromatic loci). Primers and probes are located in the first kb of the transcribed region.

Immunocytochemistry/ Immunofluorescence - Histone H3 (tri methyl K9, di methyl K9) antibody [6F12-H4] - ChIP Grade (ab71604)
ICC/IF image of ab71604 stained HeLa cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal Goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab71604, 1µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 Goat anti-Mouse IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue). This antibody also gave a positive result in 4% PFA fixed (10 min) Hek293, MCF7 and HepG2 cells at 5µg/ml, and in 100% Methanol fixed (5 min) HeLa, Hek293, HepG2 and MCF7 cells at 5µg/ml.

ELISA - Histone H3 (tri methyl K9, di methyl K9) antibody [6F12-H4] - ChIP Grade (ab71604)
ELISA using ab71604 at varying antibody concentrations. Curve_SPL7 indicates binding to the tri methyl K9 peptide ab1773. In addition, SPL6 indicates binding to the di methyl K9 peptide ab1772. Binding to the following peptides was not seen: SPL1 unmodified Histone H3, SPL2 Histone H3 mono methyl K4, SPL3 Histone H3 di methyl K4, SPL4 Histone H3 tri methyl K4, SPL5 Histone H3 mono methyl K9, SPL8 Histone H3 mono methyl K27, SPL9 Histone H3 di methyl K27, SPL10 Histone H3 tri methyl K27.
