Application Immunocytochemistry/ Immunofluorescence
Rat primary neuronal cultures where grown on cover-slips. The cells where fixed using 4% PFA. Post fixation the cells where permeabilized with 0.25% Triton-X and blocking was performed with PBS containing 3% bovine serum albumin and 5% goat serum for 1 h at 25C.

The cultures were stained with GR specific primary antibody BuGR2(1 : 200, Abcam Ab) for 1 h. After multiple washes with PBS Cultures were stained with incubation with 1 : 1000 dilution of the secondary anti-mouse IgG antibody ChromeoTM 546 (Abcam) for 1 h at 25C. The cells were finally washed with PBST (PBS with 0.01% Triton-X) and mounted after staining with DAPI nuclear stain.
