WB Use a concentration of 1 - 5 µg/ml. Detects a band of approximately 50 kDa (predicted molecular weight: 48 kDa).
ELISA Use at an assay dependent concentration. The detection limit for recombinant tagged FOXA2 is approximately 0.03ng/ml as a capture antibody.
Flow Cyt Use 0.5µg for 106 cells. ab170191-Mouse monoclonal IgG2a, is suitable for use as an isotype control with this antibody.
ICC/IF Use a concentration of 10 µg/ml.

  • 機能Transcription factor that is involved in embryonic development, establishment of tissue-specific gene expression and regulation of gene expression in differentiated tissues. Is thought to act as a 'pioneer' factor opening the compacted chromatin for other proteins through interactions with nucleosomal core histones and thereby replacing linker histones at target enhancer and/or promoter sites. Binds DNA with the consensus sequence 5'-[AC]A[AT]T[AG]TT[GT][AG][CT]T[CT]-3' (By similarity). In embryonic development is required for notochord formation. Involved in the development of multiple endoderm-derived organ systems such as the liver, pancreas and lungs; FOXA1 and FOXA2 seem to have at least in part redundant roles. Originally discribed as a transcription activator for a number of liver genes such as AFP, albumin, tyrosine aminotransferase, PEPCK, etc. Interacts with the cis-acting regulatory regions of these genes. Involved in glucose homeostasis; regulates the expression of genes important for glucose sensing in pancreatic beta-cells and glucose homeostasis. Involved in regulation of fat metabolism. Binds to fibrinogen beta promoter and is involved in IL6-induced fibrinogen beta transcriptional activation.

Anti-FOXA2 antibody 画像

  • Anti-FOXA2 antibody (ab60721) at 1 µg/ml + HepG2 cell lysate at 25 µg

    Predicted band size : 48 kDa
    Observed band size : 50 kDa (why is the actual band size different from the predicted?)
    Additional bands at : 40 kDa,42 kDa. We are unsure as to the identity of these extra bands.
  • ab60721 at 10 ug/ml staining FOXA2 in human Hella cells by Immunocytochemistry/ Immunofluorescence.

  • Overlay histogram showing HepG2 cells stained with ab60721 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab60721, 0.5µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.
