The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
Peptide ELISA: Use at an assay dependent dilution. Antibody detection limit dilution 1/16,000.
WB: Use at a concentration of 0.3 - 1 µg/ml. Detects a band of approximately 65 kDa (predicted molecular weight: 65 kDa).
Not yet tested in other applications.
Optimal dilutions/concentrations should be determined by the end user.
Expressed in the pancreas, kidney, liver, lung, placenta, brain and heart.
Belongs to the sulfatase family.
The conversion to 3-oxoalanine (also known as C-formylglycine, FGly), of a serine or cysteine residue in prokaryotes and of a cysteine residue in eukaryotes, is critical for catalytic activity.
All lanes : Anti-Arylsulfatase D antibody (ab81978) at 0.3 µg/ml
Lane 1 : A459 lysate in RIPA buffer Lane 2 : A459 lysate in RIPA buffer with immunising peptide
Lysates/proteins at 35 µg per lane.
Predicted band size: 65 kDa Observed band size: 65 kDa Additional bands at: 35 kDa. We are unsure as to the identity of these extra bands.
An additional band of unknown identity was also consistently observed at 35kDa. This band was successfully blocked by incubation with the immunising peptide.
Primary incubation was 1 hour. Detected by chemiluminescence.
