Human Prekallikrein 1B ELISA Kit (ab202405) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Prekallikrein 1B protein in cit plasma, edta plasma, hep plasma, milk, serum, and cell culture supernatant. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Prekallikrein 1B with 154 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
ASSAY SPECIFICITY This kit recognizes Prekallikrein 1B protein in serum, plasma, milk and cell culture supernatant only. Cell and tissue extract samples have not been tested with this kit
CROSS REACTIVITY Human SERPINA-5 and Human HMW Kininogen were prepared at 100 ng/mL and assayed for cross reactivity. No cross-reactivity was observed.
INTERFERENCE Human SERPINA-5 and Human HMW Kininogen were prepared at 100 ng/mL and assayed for assay interference in the presence of 10 ng/mL of Plasma Kallikrein 1B. No interference was observed.
SPECIES REACTIVITY This kit recognizes Human Prekallikrein 1B protein.
Reactivity to other species was determined by measuring 0.01% serum samples of various species, interpolating the protein concentrations from the human standard curve, and expressing the interpolated concentrations as a percentage of the protein concentration in human serum assayed at the same dilution.
Reactivity < 1% was determined for the following species: Mouse, Rat, Guinea Pig, Rabbit, dog, Sheep, Goat, Pig and Cow. -
特記事項
Prekallikrein 1B (KLKB1) is a 88 kDa serine protease involved in blood coagulation, kinin generation, fibrinolysis and inflammation. Prekallikrein 1B is synthesized in the liver and circulates in the blood stream as a free zymogen or in a complex with high molecular weight kininogen. Prekallikrein 1B has a signal peptide (aa1-19) and a pro form (aa20-638). It is composed of a heavy chain and a light chain which are cleaved upon activation.

Serum from 10 healthy male donors was measured in duplicate. The mean Plasma Kallikrein 1B concentration was determined to be 112.6 µg/mL with a range of 80.8 – 206.4 µg/mL in male donors.

Linearity of dilution is determined based on interpolated values from the standard curve. Linearity of dilution defines a sample concentration interval in which interpolated target concentrations are directly proportional to sample dilution.
Native PreKallikrein 1B was measured in the following biological samples in a 2-fold dilution series. Sample dilutions are made in Sample Diluent NS.
