Vesicular stomatitis virus (VSV), an enveloped RNA virus from the Rhabdoviridae family, is released from the plasma membrane of host cells by a process called budding. The glycoprotein (VSV-g) contains a domain in its extracellular membrane proximal stem that appears to be needed for efficient VSV budding.

ab1874 detecting bound HS scFv antibody in rat lung tissue by Immunohistochemistry (Frozen sections).

Lungs from Sprague-Dawley rats were harvested at embryonic days 17.5 of gestation and fixed with 4% (w/v) paraformaldehyde in phosphate-buffered saline (PBS), pH 7.4. The lungs were washed in PBS, cryoprotected with 20% (w/v) sucrose in PBS, and gelatin-embedded (7.5% (w/v) gelatin, 15% (w/v) sucrose in PBS) before being covered in Cryo-M-Bed and snap frozen at -40 °C. The lung sections were cut at 8 µm and mounted onto chrome alum gel slides for storage at -40 °C. Tissue sections were blocked with 10% (v/v) goat serum in PBS and incubated overnight at 4 °C with HS scFv antibody, diluted 1/5 in 1% (v/v) goat serum in PBS. Bound antibody was detected with ab1874 diluted 1/200 in 1% (v/v) goat serum in PBS, followed by fluorescein isothiocyanate-conjugated goat anti-rabbit IgG.
