Our Abpromise guarantee covers the use of ab12126 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
ICC/IF: Use a concentration of 5 µg/ml
WB: Use a concentration of 0.5 - 2 µg/ml.Detects a band of approximately 46 kDa (predicted molecular weight: 50.8 kDa).(For optimal results, primary antibody incubations should be performed at room temperature.)
TROY is a member of the TNF receptor superfamily. This receptor is highly expressed during embryonic development. It has been shown to interact with TRAF family members, and to activate the JNK signaling pathway when overexpressed in cells. This receptor is capable of inducing apoptosis by a caspase-independent mechanism, and it is thought to play an essential role in embryonic development. Alternatively spliced transcript variants encoding distinct isoforms have been described.
Type I membrane protein (Probable).
Western blot - TROY antibody (ab12126)

Predicted band size : 50.8 kDa
TROY detection by Western blot. The analysis of human TROY using this antibody in T98G, a human glioblastoma cell line extract. A protein band of approximately 46 kDa should be detected.
Immunocytochemistry/ Immunofluorescence - Anti-TROY antibody (ab12126)

ICC/IF image of ab12126 stained Mouse embryonic stem cells cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab12126, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96879 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
