iPS derived neurons (35 days) were fixed in 4% formaldehyde (10 min) and then incubated in 1% serum, 0.1% triton, 0.1% BSA in PBS for 30 mins to permeabilise the cells and block non-specific binding. The cells were then stained with antibody ab32127 (Synaptophysin at 1/250 dilution) for 1h at room temp. The secondary antibody was Donkey Anti-Rabbit IgG H&L Alexa 488 (ab150061) (green) at 1:250 dilution and incubated for 1h at room temp. Total cells were stained with DAPI (blue). Excellent positive Synaptophysin staining was observed in the processes (green).
