Additional Notes
We used immunofluoresence staining on frozen sections of the hamster ovary. The antibody seemed to recognize the STRA8 protein in fresh-frozen E13 golden hamster ovary fixed with 4% PF at 10oC. Sections were blocked and permeabilized with 10% donkey serum/0.2% Triton X100 followed by incubation with Signal Enhancer as recommended by the manufacturer. Confocal microscopy revealed low intensity STRA8 staining (red), which could be due to the level of protein expression, appreciable nuclear localization of STRA8 was noted in some germ cells (arrows). A distinct STRA8 signal was also noted in the cytoplasm, which reflected the nuclear export of the protein following the initiation of meiotic DNA synthesis. Nuclei were stained blue with DAPI.
