Thank you for your response, please see below the response from the customer; I thought I had explained myself clearly. I am well aware that these are donkey derived antibodies! I am using them against sheeppolyclonal antibodies (purified to contain onlyIgG). The sheep polyclonals are just attached to beads. The beads are then measured for PE fluorescence. The fact is that the same product made byanother company seems to work quite well whereas the Abcam one does not. I'm not sure what other information I can give, but I'm pretty disappointed with that response from Abcam. Kind regards, |

My apologies! Sorry that was pretty silly of me to forget the catalogue number .. K It is ab7009 Donkey F(ab')2 polyclonal Secondary Antibody to Sheep IgG - H&L (PE), pre-adsorbed Please see below for an email we received from a customer; I have tested this product (twice) and was disappointed with the results. When compared to a 4 year old vial of the equivalent Santa Cruz product, the Abcam antibody produced about 1/10th the signal. The application was a validation of sheep polyclonal antibody coupled beads, used for and read by a Bio-plex instrument (basically measuring the amount of sheep antibodies bound to the beads). All conditions in the experiment were the same, including test sample and antibody concentration. Lot #: GR38595 Sapphire order number: PO-13364 Abcam invoice number : 1315698B Sorry again! Thank you for response and for confirming the product code, the batch and the order number> I am a bit confused about this enquiry since ab7009 is a donkey polyclonal antibody and not sheep, so it contains donkey IgG (F(ab')2) - fragment.
It was raised against sheep IgG so the immunogne is sheep IgG whole molecule but the actual product consists of donkey immunoglobulins.
Furthermore, ab7009 has not been tested in IP or any separation technique which requires beads. It has been characterized ICC/IF, IHC-P, IHC-Fr, Immunomicroscopy, Flow Cyt.

Antibody seems not to perform recognizing serum opsonised beads or lateral flow capture lines that get a strong signal when using monoclonal anti sheep FITC antibody. Antibodies in lateral flow were also not recognised by anti-lambda ovine antibody so potentially the polyclonal recognises preferentially lambda which would be correct as sheep have 99% lambda antibodies. It would be worthwhile testing this out and put a warning onto the use of the antibody. |
