Sorry for the delay in response, I have on holidays.

The secondary antibody was originally stored at 4°C, same as the primary. I was gating on CD144 positive endothelial cells from a mixed population (gating strategy). I have had no use for the secondary with other antibodies; though could see a titration effect when titrating secondary.

Of note, the marker is listed as present on brain, though would not work with chicken brain cells.

I appreciate the time the customer has spent on these experiments and would be pleased to arrange a free of charge replacement or credit note in compensation.

On this occasion, as previously discussed I am pleased to make an exception with regards to the storage in this case. However, for future reference we would suggest to encourage the customer to store products as directed on the datasheet so that they will remain stable and we can continue to provide our guarantee. The secondary antibody has also been stored incorrectly in this case. The datasheet recommends to aliquot and store at -20oC.

I look forward to hearing from you with details of how you would like to proceed.

wenn Sie davon ausgehen, dass es eventuell eine Verunreinigung oder Beschädigung auf dem Tranportweg gab, würden ich erstmal einen Ersatz bevorzugen. Wenn der Ersatz auch wieder unspezifisch bindet/färbt, wünsche ich eine Preiserstattung.

Das Problem ist, dass es kaum einen Antikörper für das Schwein gibt. Deshalb wäre ich eigentlich auf den ab6717 angewiesen.

Ist es möglich so vorzugehen?

Dies ist absolut kein Problem, ich werde Ihnen gerne ein neues Röhrchen des ab6717 senden wenn Sie dies wünschen. Dies neue Röhrchen ist natürlich auch von unserer Garantie abgedeckt.
Ich bin mir nur nicht ganz sicher ob ich Alles richtig verstanden haben. Der ab6717 reagiert mit Kanninchen Antikörper. Dieser Antikörper wird mit all den Kanninchen- Antikörper reagieren, unabhängig davon ob diese mit Schwein oder nicht reagieren. Dieser Antikörper ist ja nicht selbst gegen das Schwein gerichtet. Falls ich dies richtig verstanden habe, ist dann auch der ab96899 geeignet diese Kanninchen anti- Schwein Antikörper zu detektieren. Ich habe Ihnen den ab96899 empfohlen, da er etwas aufgereinigter ist als der ab6717. Daher würde ich auch etwas weniger Hintergrund erwarten.

anbei finden die die Isotypkontrolle. Diese wird natürlich immer mitgeführt. Falls es positive Signale bei der Isotypkontrolle gibt, werden diese bei den Proben abgezogen, das ist auch bei den bereits gesendeten Bildern erfolgt.
Wenn grössere Mengen toter Zellen auftreten, werden diese mittels PI-Färbung ausgeschlossen.
Die Daten aus der letzten email zeigen die Ergebnisse für den Ansatz: Zellen + sekundärer Antikörper, ohne primären Antikörper.

Leider weiss ich demfall auch nicht woher dieser Hintergrund kommt. Vielleicht wurde das Röhrchen des ab6717 beim Transport oder bei der Lagerung beschädigt. Ich werde biete Ihnen auf jeden Fall entweder ein Ersatzröhrchen oder die Rückerstattung des Preises des ab6717 an. Bitte lassen Sie mich wissen, wie Sie vorgehen möchten.
