1-) Can antibody Anti-MAP2 (ABCAM - Code ab5392) be used as a marker for myenteric neurons in rat and mouse specimen?

2-) If that's possible, which one do you consider the best choice for marking myenteric neurons (total population): anti-MAP2 (ab5392) or anti-PGP9.5 (ab8189)?

1. I have done a short literature search and can confirm that the MAP2 protein is a good marker for myeneric neurons as well as neurons in general. However, I am sorry that as far as I am aware this antibody has not specifically been tested for myeneric neurons and we have no data. However, we do guarantee it for rat and mouse samples.

2 Again, PGP9.5 is a neuronal maker and soshould be a good marker for myeneric neurons. However, this has not been specifically tested. Therefore, as both antibodies have not been usedfor specificdetection of myeneric neurons Iam regrettablyunable to say which would be better.

I can suggest the customer could consider trying a further literature search for further information on which of these proteins is the better marker for myeneric neurons.

Hi,

A customer is about to purchase this antibody “Anti-PGP9.5 antibody [13C4 / I3C4] - Neuronal Marker (ab8189)” to do IHC in mouse tissue. They have had problems with anti-PGP antibodies in the past and would like to make 100% sure that this one will work. They plan to follow Abcam’s protocol for “MOUSE ON MOUSE STAINING”. They have a concern they would like clarified: The customer is concerned that in the scientific support section of Abcam’s website, in Question 3, posted Tuesday 10-February-2004, It says: “Thank you for your enquiry. Ab8189 is the only PGP9.5 antibody that we currently have and cross-reacts with Human, Rat and Guinea pig - not yet tested in other species.” The customer would like to confirm that the information in the datasheet for AB8189, which says that this antibody cross reacts with mouse, is correct and supersedes the information in the answer to Question 3 above.

You are correct in thinking that the new information of the species reactivity for this antibody:

Reacts with: Mouse, Rat, Sheep, Rabbit, Guinea pig, Dog, Human, Pig

This supersedes the prior information given as an answer to the FAQ. We have several references to this antibody having been used in staining of mouse samples, a few examples of which are given below:

I have a customer enquiry regarding ab8189. Please refer to her e-mail:

We're thinking of using this antibody on guinea pig retinal tissue, as a pan-neuronal marker. It says on the datasheet that immunofluorescence using this antibody produces high background staining. Is there another antibody that you stock which would be better? There don't seem to be many that are guaranteed to work on guinea pig tissue.

In addition to ab8189 we have three neuronal markers available that have been applied by IHC and have been shown to or predicted to cross react with guinea pig.

ab9481 is a Calbindin Mouse monoclonal antibody [CL-300] - Neuronal Marker shown to work by IF, IHC-Fr, and IHC-P.

ab11145 is a Goat Polyclonal PGP9.5 antibody - Neuronal Marker shown to work by WB and predicted to work on guinea pig based on 100% identity with the immunogen.

ab3966 is a Mouse Monoclonal 200 kD Neurofilament Heavy antibody [RNF402] - Neuronal Marker shown to work by IHC-Fr, IHC-P, and WB.

Thank you for your enquiry. Both of these antibodies (ab10358 and ab8189) have not yet been tested in Immunocytochemistry. Ab10358 has been tested in Western blot and Immunohistochemistry (Frozen sections); ab8189 has been tested in Immunohistochemistry, both formalin-fixed paraffin-embedded sections and frozen sections.
