It is possible that the antibody is not suitable for use in this application. However, since you are seeing slight staining in one of your samples, it may be possible to get this to work by optimizing your protocol.
- The incubation time of your primary antibody is rather short. We recommend incubating at least 2-4 hours at room temperature, but preferably overnight at 4C.
- Perhaps your antigen retrieval step was not extensive enough. You may wish to try different methods (longer retrieval time, different pH, enzymatic instead of heat-mediated).
