Review of Abcam Plasminogen Human ELISA Kit:
The Plasminogen ELISA, using the Abcam human ELISA kit Ab108893, Lot GR270028-6, was performed by an experienced laboratory technician at an accredited university. The goal of the experiment was to determine the cross-reactivity for rhesus macaque in both EDTA prepared plasma and in serum samples.
Overall, the kit, like other Abcam ELISA kits, was easy to follow and simple to use.
Our experiment consisted of using EDTA-prepared plasma samples and serum samples from N=3 M. mulatta. Samples were stored prior to thaw at -80 degrees Celsius. Samples were diluted serially in a range from 1:5000 to 1:80000 in the given diluent in duplicate. The rest of the assay was performed per protocol’s instructions.
At 1:80000, all samples were below the range of detection. At 1:40000, most samples were below the range of detection. At 1:20000 a few samples were below the range of detection, but most were in range. 1:5000 and 1:10000 were both in the range of detection for all sample types *subtracted ODs in the 0.6 and 0.5 range respectively). The data presented are the extrapolated values.
Overall, the kit is cross-reactive with both Rhesus plasma and Serum, however we were unable to confirm linearity of detection and value ranges are less than expected.
