Question 1

Thursday 17-August-2006

We are planning to buy your new Hsp90 antibody (ab34909) for use in an ELISA set up with Drosophila melanogaster. Have you actually tested whether this antibody works on this species in an ELISA set up (or whether other antibodies - e.g. your monoclonal Hsp90 antibody would be more appropriate)? We need to be able to detect small differences and therefore need to know about detection levels, sensitivities and how specific the antibody is. Also can you suggest secondary antibodies?

ANSWER:

Thank you for your interesting enquiry. I have looked at the details for all the antibodies we have raised against Hsp90. There is another one available, rabbit polyclonal ab13494 that will be more suitable for your application. It has been tested in ELISA and also in Drosophila.

The sensitivity of the assay you develop is going to depend on the buffers, incubation conditions, substrate, etc. All antibodies will require optimisation for your specific application and conditions. However, there is a citation for this antibody in which they used a 1:100 dilution in ELISA.

There is also a citation in which this antibody(ab13494) was used against drosophila samples.

Available secondary antibodies are: Biotinylated ab6734, ab37961 HRP ab37940, ab6734 Alk phos ab6846

I hope this helps. Please don't hesitate to get back to me if you have any more queries.
